A new avenue of hope in the fight against pancreatic cancer has emerged from research conducted in Spain. Mariano Barbacid, Director of the Experimental Oncology group at the Spanish National Cancer Research Centre (Cnio), announced findings from a study on mice demonstrating the elimination of cancerous cells associated with the most common and aggressive form of the disease, pancreatic ductal adenocarcinoma (PDAC).

The Treatment

The study’s key treatment involved a combination of three drugs targeting fundamental mechanisms that allow cancer cells to grow. Two of the drugs focused on Egfr and Stat3, proteins crucial in pancreatic cancer development, while the third targeted the oncogene Kras, considered a primary driver of the disease. Researchers reported a significant reduction in side effects alongside an unprecedented duration of tumor cell elimination.

Barbacid presented the findings alongside co-lead author Carmen Guerra, and first authors Vasiliki Liaki and Sara Barrambana, during a press conference. The study was published in ‘Pnas’ (Proceedings of the National Academy of Sciences).

Expert Insight: The challenge of drug resistance is a significant hurdle in cancer treatment. This study’s focus on overcoming resistance to Kras inhibitors represents a potentially important step forward, given that the Kras gene is mutated in 90% of pancreatic cancer patients.

Researchers at Cnio note that while new drugs blocking the action of the mutated Kras gene have been approved, their effectiveness is limited by the development of tumor resistance after several months. This new study directly addresses this issue, representing a pioneering effort in both Kras research and the development of animal models for pancreatic cancer.

Despite the promising results, Barbacid cautioned that clinical trials with the triple therapy are not yet possible. Further research is needed before this approach can be tested in human patients. It is possible that future studies will focus on refining the drug combination or identifying biomarkers to predict which patients might benefit most. Analysts expect that overcoming the challenges of translating these experimental results into clinical practice will require significant further investigation.
